The Sanger Institute Malaria Programme generates and analyses large-scale datasets in order to understand Plasmodium biology, and identify and prioritise new drug and vaccine targets. As part of this programme, the Billker and Rayner teams have founded the Plasmodium Genetic Modification Project (PlasmoGEM) to develop new scalable technologies for Plasmodium experimental genetics, and apply them to carry out genome-scale genetic screens. Using recombineering to build gene targeting vectors, and next generation sequencing to phenotype multiple knockout lines in parallel, the PlasmoGEM team has generated quantitative growth rate data for >3,000 genes in the rodent P. berghei model. Aspects of this approach are now being transferred to the major human pathogen P. falciparum.
